Because of the side effects of overdose of P4, as a multifunctional hormone, its close dosage screening without toxic reagents and complex equipment is essential from a medicolegal view. This study introduces the design and fabrication of a wearable aptasensing platform for the analysis of P4 in human biofluids, supported by a molecular dynamics (MD) simulation. The cornerstone of the proposed methodology is the use of a novel nanocomposite comprising MXene and Cu(OH)2 nanorods (Cu(OH)2NRs) to densely anchor aptamer (Apt) sequences to the flexible and low-cost graphite sheet (GS) surface in line with green chemistry principles. The aptasensor detects 1 nM to 1.5 μM of P4 with a limit of detection of 333.3 pM and a sensitivity of 0.0126 μA nM−1. The unique advantages of GS in terms of flexibility and scalability, integrated into the aptasensing interface, are considered key items for miniaturization and commercialization, aiming to promise a point-of-care test device as a home kit for non-invasive diagnosis of P4. © 2025 Elsevier B.V.
